Pengaruh Lidokain Intravena Terhadap Kadar Superoxide Dismutase I (SOD1) Dan Gambaran Histopatologi Jantung Kelinci Pada Myocardial Ischemic Reperfusion Injury Model
ABSTRACT

Background : Ischemic reperfusion injury (IRI) is the tissue damage caused by the return of blood supply to the tissue after a period of ischemia or lack of oxygen. One treatment option to prevent tissue injury is to enhance the natural antioxidants that are inside the cell such as superoxide dismutase (SOD).
Aim : Proving the effect of intravenous lidocaine on levels of superoxide dismutase 1 (SODl) and the description of a rabbit's myocardial histopathology on myocardial ischemic reperfusion injury models.

Methods : This research includes laboratoiy experimental design with post-test only controlled group. This study uses 16 local male rabbits, aged 1-2 years and weigh 1,5- 2,5 kg. The study used two groups: control group (KK) consisting of 8 rabbits were given treatment of myocardial ischemic reperfusion injury, Treatment group (KP) comprises 8 rabbits get treatment of myocardial ischemic reperfusion injury and given 2% lidocaine i.v at a dose of 1.5 mg/kg/h continuously from the beginning to the end of the procedure. SODl levels checked using the ELISA method while histopathological tissue examined using a microscope and then the result is calculated usittg linear regression equation.

Results : SODl levels p value = 0.009. for p <0.05 it can be concluded there is a significant difference between control and treatment groups. For a description of histopathology p value = 0.323, because p> 0.05 it can be concluded there is no significant difference between control and treatment groups. Conclusion: There are significant differences in the levels of SODl heart rabbit myocardial ischemic reperfusion injury given lidocaine compared to rabbits which did not receive lidocaine. As for the description of histopathology are no significant differences in myocardial ischemic reperfusion injury of rabbits given lidocaine compared to rabbits which did not receive lidocaine.

ABSTRAK

Latar Belakang: Ischemic reperfusion injury (IRI) adalah kerusakan jaringan yang disebabkan oleh kembalinya pasokan darah ke jaringan setelah periode iskemia atau kekurangan oksigen. Salah satu pilihan terapi untuk mencegah cedera jaringan adalah dengan meningkatkan antioksidan alami yang berada di dalam sel seperti superoxide dismutase (SOD).
Tujuan : Membuktikan pengaruh lidokain intravena terhadap kadar Superoxide Dismutase 1 (SOD J) dan gambaran histopatologi jantung kelinci pada myocardial ischemic reperfusion injury model.
Metode : Penelitian menggunakan 16 ekor kelinci lokal jantan, berumur 1 - 2 tahun, berat badan 1.5 - 2,5 kg. Penelitian menggunakan 2 kelompok yaitu kelompok kontrol (KK) terdiri dari 8 ekor kelinci yang diberikan perlakuan myocardial ischemic reperfusion injury. Kelompok perlakuan (KP) terdiri 8 ekor kelinci mendapatkan perlakuan myocardial ischemic reperfusion injury dan diberikan lidokain 2% i.v dengan dosis 1.5 mg/kg/jam secara kontin>ii dari awal sampai akhir prosedur. Kadar SOD1 diperiksa dengan metode EL1SA sedangkan histopatologi jaringan diperiksa menggunakan mikroskop kemudian hasilnya dihitung dengan persamaan regresi linier.

Hasil : Kadar SOD1 didapatkan nilai p = 0,009. karena p < 0.05 maka dapat disimpulkan terdapat perbedaan bermakna antara kelompok kontrol dan perlakuan. Untuk gambaran histopatologi didapatkan nilai p = 0,323, karena p >0,05 maka dapat disimpulkan terdapat perbedaan tidak bermakna antara kelompok kontrol dan perlakuan.

Simpulan : Terdapat perbedaan bermakna kadar SOD1 jantung kelinci pada myocardial ischemic reperfusion injury yang diberikan lidokain dibandingkan dengan kelinci yang tidak mendapat lidokain. Sedangkan untuk gambaran histopatologi terdapat perbedaan tidak bermakna pada kelinci myocardial ischemic reperfusion injwy yang diberikan lidokain dibandingkan dengan kelinci yang tidak mendapat lidokain.
